Objectives Essential hypertension (EH) is an etiological risk factor that poses a serious threat to human health. The aim of this study therefore was to investigate the correlation between the two SNPs (rs1805067 and rs2397771) of the norepinephrine (NE) transporter gene in the sympathetic nervous system and essential hypertension (EH). Methods A total of 96 hypertensive patients (EH group) and 96 normal subjects (control group) were selected by epidemiological investigation and the sequences at rs1805067 and rs2397771 in the two groups were investigated using polymerase chain reaction (PCR) and Sanger sequencing. In the EH group, systolic and diastolic blood pressure, body mass index (BMI), levels of creatinine, low-density lipoprotein cholesterol (LDL-C), triglycerides (TG), and the percentage of drinkers were found to be higher compared with those of the control group (P < 0.05). e112

Results The results of the analysis of the allele frequencies of rs1805067 (A/G) revealed that there was no difference between the two groups (P > 0.05). The analysis results of the allele frequencies of rs2397771 (C/G) in the EH group revealed that G accounted for 48.96% and C for 51.04% of the patients, while in the control group, these values were 64.58 and 35.42%, respectively. Conclusions These results indicated a significant difference between the two groups (P < 0.05). In the population studied, the onset of EH may be correlated with BMI, TG, LDL-C and high-density lipoprotein cholesterol (HDL-C). EH may be related to the rs2397771 (C/G) C allele polymorphism of the NE transporter gene but not to rs1805067 (A/G).

GW24-e1783 THE EXPERIMENTAL STUDY OF PROTECTIVE EFFECTS OF ATRIAL NATRIURETIC PEPTIDE (ANP) INJECTION ON THE CULTURED HUMAN UMBILIAL VEIN ENDOTHELIAL CELLS INJURED BY HYPOXIAREOXYGENATION Chen Zhang Qiang. Jiangxi Provincial People’s Hospital 10.1136/heartjnl-2013-304613.306

Objectives To study the protective effects of Atrial Natriuretic Peptide (ANP) Injection on the cultured human umbilial vein endothelial cells injured by hypoxia-reoxygenation Methods hypoxia-reoxygenation group (n = 6): cells were cultured in the hypoxia box for 6 hours, then reoxygenation for 6h; 3. hypoxia-reoxygenation + different concentrations of ANP Group: 0.001 µg/ml (n = 6); 0.005 µg/ml (n = 6); 0.01 µg/ml (n = 6); 0.05µg/ml (n = 6); 0.1µg/ml (n = 6), and also hypoxia cultured for 1 h and reoxygenation 6 h; the cells in the cell morphology was observed under inverted microscope, cell culture supernatant obtained in each group were measured {malondialdehyde (MDA), lactate dehydrogenase (LDH), nitric oxide (NO) and endothelin (ET) content}, the final results of statistical analysis conducted. Results The dehydrogenase (LDH) activity in hypoxia-reoxygenation group was significantly higher compared with the control group (69.35 ± 5.66 vs 31.04 ± 3.43, P < 0.01); LDH activity in drug intervention group was significantly decreased compared with hypoxic reoxygenation (P < 0.01). MDA content in hypoxia-reoxygenation group was significantly higher than that in the control group (5.94 ± 0.58 vs1.69 ± 0.16, P < 0.01), but MDA content in drug intervention group was significantly lower than that in hypoxia-reoxygenation group (P < 0.01), during the range of ANP concentration from 0.001 µg/ml to 0.05 µg/ml), MDA content in the culture medium concentration showed negative correlation with ANP concentration in the ANP group (P < 0.01). Levels of intracellular SOD in hypoxia-reoxygenation group were significantly lower than that in the control group (15.74 ± 2.17 vs 47.08 ± 4.23, P < 0.01) and was significantly higher in drug treatment group than in hypoxia-reoxygenation group (P < 0.01), When ANP concentration range from 0.001 µg/ml to 0.05 µg/ml, each sub-group correlation analysis showed medium concentration of SOD content was positively correlated with ANP concentration in ANP intervention group (P < 0.01); Nitric oxide (NO) levels in cell culture medium in hypoxia group was significantly lower than that in the control group (36.81±3.78 vs 89.78±7.01, P 0.05). RSR revealed that the reaction system of PON activity and Ca2 + significantly contributed to the levels of LDL-HNE (P < 0.05). However, the interaction of Ca2+significantly contributed to the CHD with normal serum lipid levels (P < 0.05). Conclusions H2O2, systolic pressure, hypersensitive c-reactive protein contributed to the CHD with normal serum lipid levels. The reaction system of PON activity and Ca2+ contributed to the levels of LDL-HNE. However, the interaction of Ca2+conntributed to the CHD with normal serum lipid levels.

A polarizable dipole-dipole interaction model for evaluation of the interaction energies for N-H···O=C and C-H···O=C hydrogen-bonded complexes.

In this article, a polarizable dipole-dipole interaction model is established to estimate the equilibrium hydrogen bond distances and the interaction ...
