Complete nucleotide sequence of Friend murine leukemia'. virus, strain FB29 Sylvia Perryman, Jane Nishio and Bruce Chesebro Laboratory of Persistent Viral Diseases, Rocky Mountain Laboratories, National Institute of Allergy and Infectious Diseases, Hamilton, MT 59840, USA Submitted November 12, 1991 Friend ecotropic murine leukemia virus (F-MuLV) isolate I-s derived from long-term bone marrow cultures which had been infected by Friend virus polycythemia-inducing virus complex (FV-P) by Mathieu-Mahul et al. (1). The F-MuLV strain FB29, derived from the I-5 isolate by Sitbon et al., exerts cytolytic and leukemogenic effects in erythroid cells, leading to a severe early hemolytic anemia (EHA) followed by a late erythroleukemia in newborn-inoculated susceptible mice (2). Latency of induction of erythroleukemia has been mapped to the U3 region of the viral long terminal repeat (2, 3). The major determinant of the severe EHA mapped to the envelope gene of FB29 (2). However, with experiments using another virulent strain of F-MuLV, 57, it was shown that variations in three different regions can influence the severity of the EHA (3, 4). These included a conserved structural envelope determinant, transcriptional enhancer sequences in the U3 region, and sequences in the U5-gag-pol regions. The F-MuLV FB29 clone containing the entire viral genome was ligated into the vector pBR322 at the HindIII site within the pol gene (2) (base 5060 of the given sequence). Sequence data was obtained on double-stranded plasmid DNA using the dideoxy chain-termination method with E. coli DNA polymerase, Klenow fragment (BRL), and labeled with at-35S-dATP or a-32P-dATP (New England Nuclear). Some sequences were confirmed using T7 DNA polymerase (Promega) or Bst DNA polymerase (BioRad). Specific primers for sequencing were made on DNA synthesizers from Applied Biosystems (model 380B) or Biosearch (model SAM ONE, or model 8600). The sequence of the env gene of F-MuLV strain 57 was previously published by Koch et al. (5, 6, 7). The sequence of the entire genome of F-MuLV 57 is available in the EMBL data library (8). The level of homology between F-MuLV FB29 and two other closely related murine leukemia viruses, F-MuLV 57 and Maloney MuLV, is presented in the table below. was
