xanthene dyes T 1000 Fluorescent Dye Phosphoramidite Labelling of Oligonucleotides. 02 - 246 — 5-Carboxyfluorescein (I) is activated, cyclized, and finally coupled with the aminopropoxymethyldioxolane (III), forming the adduct (IV). Subsequent hydrolysis, dimethoxytritylation, and phosphitylation afford the 5-carboxyfluorescein phosphoramidite (VII). This is used in DNA synthesizers to prepare directly fluorescent dye labelled oligonucleotides (FDLO) by coupling to the 5’-hydroxylic function of the support bounded oligonucleotide.
