ADP and ionophore A33 187 were markedly reduced in platelcts from strokc-prone spontaneously hypertensive rats (SHRSP) compared with thosc ocagc-matched Wistar Kyoto rat (WKY) platelets, suggesting defeetive functions of intraccllular Ca?+ in SHRSP platelets (Tomita et al. Hypertension 19x9: I-l: iO3-315). To clarify tbe mechanism of the platelet hypofunctilons, saponin pcrmeabili/.cd platelets were prepared to compare the responses of platelct\ (‘ram both rats in varying concentrations of extracellular Ca?+. The Icakagc of laclate dehydrogenase from saponin (IS pg/ml)-treated platelets was approx. 5 !; of total activity: the dcgree of the leakage in both platelets did nol diffcr. In sqonin-trcated plstelets, extracellular Ca?+ alone did not induce either or sccretion in both strains. Howevcr, in the presence of 1-oleoyl-zagg7 ~.x
