Foreword This symposium on Toxicologic Pathology of the Upper Respiratory System is a continuation of an ongoing series of annual symposia designed to provide a forum for exchanging ideas and information on selected areas of toxicologic pathology. The symposium was organized and conducted by the Chemical Pathology Branch of the National Tbxicology Program, in collaboration with the Chemical Industry Institute of Toxicology. The purpose of the meeting was to provide a thorough documentation of spontaneous and treatment-induced lesions in the upper respiratory system of laboratory rodents. This collection of papers reflects the contemporary state of knowledge, with respect to toxicologic pathology of the nose and major airways of rats and mice used in chemical toxicity and carcinogenicity studies. Following an overview of the practical aspects and approaches to studying chemicals administered by the inhalation route, other formal presentations provide background infonnation on comparative anatomy, physiology, airflow dynamics, biochemistry, and enzyme histochemistry of the nasal cavity. Several papers discuss the

nasal pathology of rats and mice, focusing on classification and interpretation of nonneoplastic and neoplastic lesions. Comparative nasal pathology and toxicity are also addressed along with in vivo and in vitro research studies of normal and transformed respiratory epithelium. Consensus opinions of the symposium participants regarding selected histologic material that was discussed prior to the symposium are summarized, along with photomicrographs of representative lesions. Taken together, these papers represent a compilation of current knowledge relative to normal structure and function of the upper respiratory system of rats and mice, with a comprehensive characterization of infectious, toxic, and proliferative lesions. We hope that these collected papers provide a useful resource for scientists interested in defining toxic responses in the upper respiratory system.
