The Synergism of y-Interferon and Tumor Factor in Whole Body Hyperthermia

Abstract - It is suggested that y-interferon (y-IFN) can be added to tumor necrosis factor (TNF) to improve therapy in whole body hyperthermia (WBH). The amount of r-HTNF is about 3.5 x 106 units per week with about 3.5 x 104 units of rH y-INF intravenously. The body temperature should be 40°C for one half hour. An i.p. injection of secobarbital (24 mglkg) should be given as well.

Introduction Previously (l), I suggested the use of 3.5 x lo6 units of r-HTNF with whole body hyperthermia of 40°C for one half hour as a cancer therapy. This is based on (2). However, the tumors in (2) are subdermal. I am interested in eliminating metastases after sugery instead of chemotherapy.

hyperthermia can minimize the toxicity of TNF. The use of TNF in resistant cells ME- 180neo(5) can cause growth of tumor cells as well as have little effect. This depends on the number of units of r-H TNF used. If one uses rH y-TNF of approximately 10-Zof the units of r-H TNF, the resistant cells are completely destroyed. Conclusion

Side effects of tumor necrosis factor

According to Old (3) TNF can be toxic. Also, TNF may be ineffective against resistant cells. In addition (4), TNF can even enhance the growth of tumor cells.

For metastatic cancer, after surgery one should be able to control or cure patients with body hyperthermia of 40°C for one half hour upon adding 3.5 x 106 units of r-H TNF with 3.5 x lo+4 units of rH y- TNF per week i.v. In addition 24 mg/kg of secobarbital is used i.p. The exact protocol must be obtained experimentally.
