The complete mitochondrial DNA sequence of Thymallus grubii (Salmonidae: Thymallinae) was determined in this study. The complete mtDNA sequence of T. grubii was 16,657 bp in length, consisted of 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes and 1 control region. Intergenic spacer and overlapping sequences were found in mtDNA of T. grubii. The base composition of the whole mtDNA was 27.9% A, 27.2% T, 27.5% C and 17.4% G, with a slightly A þ T bias of 55.1%. Seven protein-coding genes had an incomplete stop codon in T. grubii.

The Amur grayling, Thymallus grubii, is an important commercial cold freshwater fish and mainly distributed in the Amur basin and limited to the upper levels of some rivers in China (Ma et al., 2012). Nowadays, mitochondrial DNA (mtDNA) has been widely used for molecular identification and phylogenetic analysis of closely related species. Up to date, only two Thymallus complete mtDNA had been sequenced: Thymallus thymallus (NC_012928.1) and Thymallus arcticus (NC_012929.1). However, there was no data of the complete mitochondrial genome of Thymallus grubii. In this study, the whole mitochondrial genome sequence of T. grubii was characterized using polymerase chain reaction (PCR)-based method (GenBank accession number: KF692113). The genes were annotated using MitoFish online version available at http://mitofish.aori.utokyo.ac.jp/annotation/input.html (Iwasaki et al., 2013). The sequence alignment was performed by Clustal Omega (Sievers et al., 2011). The complete mtDNA sequences of Thymallus grubii was a circular molecule of 16,657 bp in length, including

13 protein-coding genes, 22 tRNA genes, 2 rRNA genes (12S rRNA and 16S rRNA) and 1 control region (D-loop). Its organization and arrangement was presented in Table 1, which was identical to the other fish mitochondrial genomes (Hwang et al., 2013; Liang et al., 2012; Wang et al., 2013; Yasuike et al., 2010). The base composition of the whole mtDNA was 27.9% A, 27.2% T, 27.5% C and 17.4% G, with a slightly A þ T bias of 55.1%. All protein-coding genes used ATG as the start codon with exception of CO1 which used GTG. Five protein-coding genes terminated with the complete stop codon TAA (ND1, CO1, ATPase8, ND4L and ND5), ND6 regarded TAG as stop codon and the rest showed an incomplete stop codon (T or TA), which was quite common among other fishes (Hwang et al., 2013; Liang et al., 2012; Wang et al., 2013). Some overlapping sequences were found among the 13 protein-coding genes: ATPase8 gene overlapped with ATPase6 gene for 10 bp, ND4L gene overlapped with ND4 gene for 7 bp and ND6 gene overlapped with ND5 gene for 4 bp. On the other hand, some intergenic spacers were found in T. grubii mtDNA. The 22 tRNA genes ranged in length from 67 bp (tRNA-Cys) to 75 bp (tRNA-Leu), and were predicted to fold into expected cloverleaf secondary structures. The 12S and 16S rRNA genes were 947 and 1679 bp, respectively, located between the tRNA-Phe and tRNA-Leu (UUA) genes and separated by the tRNA-Val gene. The control region of T. grubii was 997 bp in length and lied between the tRNA-Pro and tRNA-Phe genes. The origin of light-strand replication was identified between tRNA-Asn and tRNA-Cys genes and was 34 bp in length. The sequence alignment showed that the mtDNA of T. grubii shared 94.71 and 94.47% sequence identity with that of T. arcticus (NC_012929.1) and T. thymallus (NC_012928.1), respectively.
